Deficient resistance to Coccidioides immitis following intravenous vaccination

II. Evidence against an immune tolerance mechanism

Abstract

Intramuscular vaccination with killed spherules induced strong immunity to lethal coccidioidal disease in mice. A wide range of dose and small incremental dosage gave uniformly poor protection when administered intravenously. In imuran- or cortisone-treated leukopenic or lymphopenic mice intravenous vaccination did not result in augmented immunity later, after leukocyte numbers had become normal. In both leukopenic and nonleukopenic intravenously-vaccinated animals the cellular response was primarily neutrophilic. These findings did not support earlier speculation that the deficient immune response in intravenously vaccinated mice was attributable to immune tolerance.

Zusammenfassung

Die intramuskuläre Impfung mit abgetöteten Sphärulen induzierte bei Mäusen eine kräftige Immunität gegen ttdliche Coccidioidomykose. Ein weitgespannter Dosisbereich und eine geringe, ansteigende Dosierung ergaben einheitlich kümmerlichen Schutz, wenn die Gaben intravenös erfolgten. In Immuranoder Cortisonbehandelten leuko-oder lymphopenischen Mäusen bewirkte die intravenöse Impfung keine spätere vermehrte Immunität, nachdem sich die Leukozytenzahlen normalisiert hatten. In leukopenischen und nicht-leukopenischen intravenös geimpften Tieren war die zelluläre Antwort meist neutrophil. Diese Befunde stützten frühere Vermutungen, dad die mangelnde Immunantwort bei intravenös geimpften Mäusen auf einer Imuntoleranz beruhten, nicht.
