Deficient resistance to Coccidioides immitis following intravenous vaccination

III. humoral and cellular responses to intravenous and intramuscular doses

Abstract

Serologic studies showed that mice vaccinated intravenously with killed Coccidioides immitis spherules developed higher precipitin titers than mice vaccinated intramuscularly. Nevertheless, the intravenously-vaccinated animals were poorly protected against challenging doses of arthrospores whereas the intramuscularly-vaccinated mice were strongly resistant. Resistance induced intramuscularly was associated with cellular immunity manifested by high leukocytolytic and macrophage aggregation indices in the presence of spherule coccidioidin. Relatedly, these mice showed stronger delayed hypersensitivity reactions than did animals vaccinated intravenously. The latter route was non-efficacious in conferring strong leukocytolytic and macrophage aggregation properties on white blood cells. The cell-mediated immune reactions depended in part on heat-labile antigen(s). The data suggested that vaccination by the intravenous route stimulated humoral immunity to the detriment of cell-mediated immunity.

Zusammenfassung

Serologische Studien ergaben, daß intravenös mit abgetöteten Coccidioides immitis-Sphärulen geimpfte Mäuse höhere Präzipitionstiter entwickelten als intramuskulär geimpfte. Trotzdem waren die intravenös geimpften Tiere schlechter gegen eine Provokationsdosis von Arthrosporen geschützt, während die intramuskulär geimpften Mäuse hoch resistent waren. Die intramuskulär induzierte Resistenz war mit zellulärer Immunität gekoppelt, angezeigt durch hohe leukocytolytische und Makrophagen-Ansammlungs-Indices in Gegenwart von Sphärulen-Coccidioidin. Diese Mäuse zeigten übrigens eine stärkere Uberempfindlichkeit vom verzögerten Typ als intravenös geimpfte Tiere. Letztgenannter Weg war nicht imstande, auf weiße Blutzellen starke leukocytolytische und Makrophagen-Aggregations-Eigenschaften zu übertragen. Die zellulären Immunreaktionen hingen teilweise von hitzelabilem Antigen (bzw. Antigenen) ab. Die Befunde lassen den Schluß zu, daß die intravenöse Impfung eine humorale Immunität auf Kosten der zellulären herbeiführt.
