A comparative study of ribonucleic acid species in yeast-like and mycelial forms of Paracoccidioides brasiliensis

Abstract

A method is described for the isolation of undergraded RNA from yeast-like (Y) and mycelial (M) forms of Paracoccidioides brasiliensis. The method is a combination of various known techniques.

Cosedimentation in a sucrose gradient of Y form RNA labelled with 14C-uracil and M form RNA labelled with 3H-uracil showed identical sedimentation behaviour of the 3 major types of RNA. However, one of these types of RNA (light ribosomal RNA) is comparatively more abundant in the M form than in the Y form of P. brasiliensis.

In addition to the 3 major sedimenting types of RNA, many other intermediate species of RNA were detected in both Y and M forms by electrophoretic analyses in polyacrylamide gel. A two-band difference was found between electrophoretic patterns of RNA sedimentation samples obtained from Y and M forms.

Resumen

Se describe un método para el aislamiento de RNA no degradado a partir de las formas levaduriforme (Y) y micelial (M) del Paracoccidioides brasiliensis. El método es una combinación de varias técnicas ya establecidas. La cosedimentación en un gradiente de sacrosa del RNA de la forma Y marcada con 3H-uracilo y del RNA de la forma M con 14C-uracilo, demostró que sus tres tipos principales de RNA tienen un comportamiento de sedimentación idéntico. Sin embargo, uno de estos tres tipos de RNA (el RNA ribosomal liviano) es comparativamente más abundante en la forma M que en la forma Y del P. brasiliensis. Además de los tres tipos principales de RNA obtenidos por ultracentrifugación, se detectaron muchas otras especies de RNA en ambas formas del P. brasiliensis mediante análisis electroforéticos en gel de poliacrilamida. La comparación de los patrones electroforéticos obtenidos de las muestras de RNA reveló en cada una de las dos formas del P. brasiliensis la presencia de una banda distintiva que no tiene homólogo en la otra forma.
