Abstract
Coccidioides immitis spherules cultured serially in a defined medium respond to certain stimuli by reverting to mycelium. A number of these stimuli have been identified and the response measured. Temperatures of incubation between 26°C and 40°C had only a minor effect on reversion. Reduction of the CO2 tension resulted in increased reversion. Human serum provided a powerful stimulus and most of the activity resided in the non-ultrafilterable fraction. No single fraction of the serum proteins was responsible for all the activity and the activity was not reversed by the addition of iron. Among saturated fatty acids maximal reversion occurred with myristic acid and maximal inhibition of growth with lauric acid. The unsaturated C18 acids were effective with reversion activity and toxicity increasing with unsaturation. Amphotericin B was both a growth inhibitor and a reversion stimulus.
Resumen
Esférulas de Coccidioides immitis cultivadas serialmente en un medio definido responden a ciertos estímulos revertiendo a la estructura miceliar. Un número de estos estímulos han sido estudiados. Las temperaturas de incubación entre 26°C y 40°C tuvieron poco efecto en la reversión. La reducción de la tensión del bióxido de carbono determinó un aumento de reversión. El suero humano fué un poderoso estímulo y la mayor parte de la actividad residía en la fracción que no era ultrafiltrable. Ninguna fracción en particular de las proteinas séricas era responsible de toda la actividad y dicha actividad no era afectada por la adicción de hierro. Entre los ácidos grasos saturados, la máxima reversión occurría con el ácido mirístico y la máxima inhibición del crecimiento con el ácido laúrico. Los ácidos no saturados C18 eran efectivos con la actividad reversible y se aumentaba la toxicidad con relación al grado de insaturación. La amfotericina B fue tanto un inhibidor del crecimiento como un estimulador de la reversión.
