Abstract
The filamentous fungus Trichophyton mentagrophytes was grown in [U-14C] glucose supplemented nutrient broth. Growth was monitored in 200 μl microcultures and 5 ml macrocultures by measuring the incorporation of 14C into trichloroacetic acid-insoluble macromolecules and in macrocultures by dry weight determination. Adjustment of the specific activity of the [U-14C] glucose medium supplement from 1080 μCi/mmole to 108 μCi/mmole decreased the rate of isotope uptake and prolonged availability so that growth beyond 36h could be monitored radiometrically. The dry mycelial weight of the fungus was directly proportional (r=0·995) to the amount of isotope incorporated. Isotope uptake in microcultures and macrocultures was kinetically identical. These data clearly indicate that the uptake of 14C by T. mentagraphytes can be used to monitor fungal growth accurately in both macrocultures and microcultures.
Zusammenfassung
Der fadenbildende Pilz Trichophyton mentagrophytes wurde in einer mit [U-14C] Glukose enthaltenden Nährlösung gezüchtet. Das Wachstum wurde radiometrisch in 200 μl Mikrokulturen und 5 ml Makrokulturen überprüft, indem die Aufnahme des Isotops in die Makromoleküle der trichloressigsäureunlöslichen Zellwand gemessen wurde. Bei den Makrokulturen erfolgte die Bestimmung auch des Trockengewichtes. Eine Änderung der spezifisehen Aktivität des [U-14C] Glukosezusatzes von 1080 μCi/mmol auf 108 μCi/mmol verminderte die Geschwindigkeit der Isotopenaufnahme und verlängerte die Verfügbarkeit, so daß das Wachstum auch über 36 Stunden hinaus radiometrisch bestimmt werden konnte. Das Trockenmyzelgewicht des Pilzes verjielt sich direkt proportional (r=0·995) zu der aufgenommenen Isotopenmenge. Die Isotopenaufnahme in den Mikro- und Makrokulturen war kinetisch identisch. Diese Befunde lassen klar erkennen, daß die Aufnahme von [U-14C] Glukose durch das T. mentagrophytes herangezogen werden kann, um radiometrisch das Pilzwachstum sowohl in Makrokulturen als auch in Mikrokulturen genau zu bestimmen.
