Scalable Synthetic Route to 2-Amino-5-hydroxypropiophenone: Efficient Formal Synthesis of Irinotecan

Abstract

2-Amino-5-hydroxypropiophenone, a key precursor in the total synthesis of irinotecan, has been synthesized. Regioselective nitration and a SNAr displacement are the key steps involved in this strategy, which is high yielding, is economical, and has been performed on a plant scale.
