Vanadium-substituted Keggin structured polyoxometalate combined with fluconazole in α-SiW11V-FKZ for efficient bacterial inhibition

Abstract

Pollution of the environment is becoming more and more serious due to the misuse of antibiotics. To protect the environment, polyoxometalates (POMs) emerged as potential green materials. These POMs contain anionic clusters of metal oxides, which exhibit favorable structural and physicochemical properties. They have demonstrated promising antiviral, antibacterial and antitumor activities in biomedicine. In the present study, fluconazole (FKZ)-modified POMs of Keggin structure were synthesized as an efficient bacteriostatic agent by the hydrothermal method. Furthermore, the bacteriostatic properties of the agent against various bacteria are analyzed. The bacteriostatic effect and bacterial inhibition rate are evaluated using three different methods: the filter paper slice method, determination of MIC value and the colony counting method. In addition, the leakage of morphologically repressive DNA of bacteria after being treated by the samples was investigated by TEM and UV, which resulted in speculation of repressive mechanisms. The experimental results demonstrated that the bacteriostat exhibited superior bacteriostatic properties compared to the classical polyoxometalate. Additionally, bacteriostatic agents are particularly effective in combating gram-negative bacteria.
