Quantitative Immunochemical Determination of Lysozyme (Muramidase) in Serum and Urine

Abstract

Immunochemical determination of human lysozyme was performed by electrophoresis of samples into antibody-containing agarose gel. Lysozyme was isolated from urine of patients with monocytic leukemia by a simple procedure involving batch adsorption of the lysozyme to CM-Sephadex. This preparation was used as standard and for the production of antisera. Normal serum lysozyme was 2.8 ± 0.8 mg/l, whereas the concentration in normal urine was below the limit of detection (0.3 mg/l). Four patients with monocytic leukemia showed large elevations of lysozyme in serum and urine. Comparison of immunochemical determination and bacteriolytic assay showed good concordance.
