EXTRACTION AND DETERMINATION OF BIOMARKERS IN FICUS CARICA L. LEAVES FROM VARIOUS SPECIES AND DIFFERENT CULTIVARS BY HPLC

Abstract

An HPLC method has been developed and validated for determination of three biomarkers, rutin, psoralen, and bergapten, in Ficus carica leaf extract. Chromatographic separation was achieved on a C18 column using A: methanol and B: ACN–water (15:85, v/v; pH 3) as the mobile phase in a gradient elution of 0–25 min from 20 to 90% of solvent A. The UV detector was set at 310 nm. The calibration curve was linear over the range of 2–700 mg/L, 0.12–150 mg/L, and 0.28–100 mg/L, for rutin, psoralen, and bergapten, respectively. Recoveries within the range of 98.1–107.8% for the components were studied. The RSD (%) was less than 7.0%. Rutin, psoralen, and bergapten were extracted from F. carica leaves by SLE. The effect of various parameters, that is, solvent, time, and pH on yield was investigated. Solvent system containing ACN:water (70:30, v/v) at pH 3–5 for 2 hr was more efficient than the other tested variables for the extraction. This HPLC assay is a precise and reliable method for the simultaneous analysis of rutin, psoralen, and bergapten in the F. carica leaf extract and was applied for different cultivars.
