Human umbilical cord-derived mesenchymal stem cells conditioned medium ameliorates CCl4-induced liver fibrosis through regulation of expression and activity of liver lysyl oxidase

Abstract

The effects of human umbilical cord blood mesenchymal stem cells conditioned medium (hMSC-CM) on Lysyl oxidase (LOX) family gene expression and proteins were evaluated. Fifty-four male Wistar rats were divided into nine groups. The control groups received olive oil and phosphate-buffered saline (PBS) while untreated groups received CCl4. The treated groups received hMSC-CM after the 4th week of CCl4 injection. LOX family expression and proteins were evaluated by RT-PCR and western blot. CCl4 enhanced significantly the expression of LOX genes and proteins. hMSC-CM significantly decreased the expression of the LOX genes and proteins. hMSC-CM decreased cross-linked collagen in the treated groups. hMSC-CM improved liver fibrosis through decreased LOX family gene expression.
