Primary subcultures of Histoplasma capsulatum

I. Macro and micro-morphology of the mycelial phase

Abstract

The colonial and micromorphology of 36 primary subcultures of H. capsulatum from various sources have been studied in depth. Most strains separate into 2 main types on the basis of colony appearance. The “A” or albino type with white aerial mycelium and smooth macroconidia and the “B” or brown type consisting almost solely of masses of tuberculate macroconidia. The implications of this in identification and epidemiology are discussed. Several new or improved cytological observations are reported including racket hyphae, nuclei and nucleoli, gelatinous wall sculptures and perforate septa.

Resumen

La observación microscopica y las caracteristicas culturales de 36 aislamientos primarios de H. capsulatum, provenientes de varias fuentes fueron estudiados. La mayor parte de las cepas se agrupan en 2 tipos fundamentales sobre las base de la apariencia en cultivo. El tipo “A” or tipo albino con micelio blanco aereo y macroconidios lisos y el tipo “B” or tipo pardo consistente casi exclusivamente en masas de macroconidios tuberculados. Las implicaciones de esto en identificacíon y epidemologia son discutadas. Algunas observaciones citologicas nuevas o meyoradas son expuestas, incluzendo hifas en raqueta, nucleos, nucleolos, extructuras gelatinosas de los paredes y divisiones de las mismas.
