Quantitative Analysis of Biogenic Amines by High-Performance Thin-Layer Chromatography Utilizing a Fibre Optic Fluorescence Detector

Abstract

The biogenic amines, putrescine, cadaverine, spermidine and spermine, dansylated in a microwave oven, were separated by HPTLC, with chloroform:triethylamine (2:1) (v:v) as eluent and silica-gel as stationary phase. A fibre optic-based fluorescence instrument for in situ scanning was used for quantitative measurements. The compounds were determined over the range 2-85 ng, with relative standard deviations between 0.75 – 1.39% and detection limits over the range 1.8 – 3.0 ng. The method was applied for the determination of biogenic amines in plant tissue.
