Diastereomeric Analysis of Bioactive N-Phenylpyrazole-4-acylhydrazone Derivatives by High Resolution Gas Chromatography

Abstract

Bioactive acylhydrazone samples prepared by condensation of 1-phenyl-4-acylhydrazide pyrazole with benzaldehyde were analyzed by 1H NMR, mass spectrometry and High Resolution Gas Chromatography (HRGC), utilizing a glass capillary column coated with SE-54 (1%vinyl-5%phenyl-94%methylpolysiloxane). HRGC was shown to be a valuable tool for the routine analysis of the acylhydrazones, and determination of the diastereomeric ratio, without need of derivatization and purification. As an example, HRGC was employed to follow the kinetics of formation at the imino double bond level, of both diastereomers of hidrazone (1a) during a synthetic procedure.
