Nickel Carbonyl

Effects Upon the Ultrastructure of Hepatic Parenchymal Cells

Abstract

The acute pathologic reactions of hepatic parenchymal cells following exposure to nickel carbonyl (Ni[CO]4) were investigated by electron microscopy. Nickel carbonyl was administered intravenously to rats in LD50 and LD100 dosages. Diffuse dilatation of rough endoplasmic reticulum was the most prominent and most consistent ultrastructural abnormality. The mitochondria and other cytoplasmic organelles were essentially unaffected by the injection of Ni(CO)4. Nucleolar alterations were present in hepatocytes during the period from 2 to 24 hours after injection. The affected nucleoli appeared to be smaller than normal, with eccentric localization of dense osmiophilic regions.
