ABSTRACT
We describe here a simple and rapid detection of methamphetamine (MA) and amphetamine (AP) simultaneously in urine by gas chromatography (GC) using a solid-phase micro-extraction (SPME) technique. We used an SPME fiber (Polydimethylsiloxane thickness; 100μM) with a fiber holder for manual use. After addition of urine (1.0 ml), NaCI(0.5g) and 1-phenylbutylamine (5.0 μg, 1-PB, internal standard) into a vial (1.5 ml), we adjusted the sample to pH12 with NaOH (10N) and then rapidly sealed it with a teflon-coated septum. After immersion of the SPME fiber in the sample for 20 min, and then washing promptly in NaOH-H3BO4 buffer (pH12), the SPME needle was inserted into the injection port of the GC and the fiber extruded. Each peak of MA, AP and 1-PB was clearly separated. Linearity of AP and MA were in the ranges of 0.25–10 and 0.1–1.0 μg/ml, respectively. This SPME technique, applicable without using organic solvents, is simple and rapid. It is, therefore, useful for the practical detection of MA and AP in urine.

Une technique de micro-extraction à phase solide (“SPME”) simple et rapide pour la détection simultanée de la méthamphétamine (MA) et de l'amphétamine (AP) dans l'urine par Chromatographie en phase gazeuse est présentée. Une fibre SPME (épaisseur de type polydiméthylsiloxane; 100 μM) ainsi qu'un support à fibre pour usage manuel ont été utilisés. Après l'addition de l'urine (1.0 mL), du NaCI (0.5g) et du 1-phénylbutylamine (5.0 μg, 1-PB, étalon interne) dans une fiole de 1.5 mL, le pH a été ajusté à 12 avec du NaOH (10N) et la fiole rapidement scellée avec un septum enduit de téflon. Après l'immersion de la fibre SPME dans l'échantillon pour une durée de 20 minutes et un lavage rapide dans un tampon NaOH-H3BO4 (pH 12), la fibre a été exposée au port d'injection du chromatographe en phase gazeuse après l'insertion de l'aiguille SPME. Chacun des pics (MA, AP et 1-PB) était clairement séparé. La linéarité de quantification de l'AP et de la MA était dans les écarts de 0.25–10 et de 0.1–10 μg/mL respectivement. Cette technique SPME qui ne nécessite aucun solvent organique est simple et rapide et donc utile pour la détection de la MA et de l'AP dans l'urine.
