ABSTRACT
The use of membrane test kits for detecting prostate specific antigen (PSA) during the forensic identification of semen has become a routine procedure in forensic labs across North America. During the validation of the “Seratec® PSA Semiquant” membrane test kit (Gesellschaft für Biotechnologie, Germany) at The Centre of Forensic Sciences, the sensitivity of the kits was found to be approximately 100 times greater than crossed-over electrophoresis. It also approached the reported sensitivity for detecting PSA through the enzyme-linked immunosorbent (ELISA) assay. As with any validation study of a new test procedure, it is a requirement that the specificity of that procedure be determined. It was during this aspect of our validation study that we obtained a positive PSA result from semen-free vaginal samples and also with a commercially available contraceptive foam. The validation results and interpretation guidelines for when the kits are used at the Centre of Forensic Sciences are outlined.
RÉSUMÉ
L'utilisation de tests membranaires pour détecter l'antigène spécifique de la prostate (PSA) lors de I'identification de sperme est devenue une procédure de routine pour les laboratoires judiciaires à travers l'Amérique du Nord. Le test membranaire suivant: “Seratec® PSA Semiquant” (Gesellschaft für Biotechnologie, Germany) fut validé au Centre des sciences judiciaires. La sensibilité de ces tests s'est avérée approximativement 100 fois plus grande que par électrophorèse (technique “cross-over”). Cette sensibilité se rapproche également de celle des tests ELISA (enzyme-linked immunosorbent assay) pour détecter l'antigène PSA. Comme pour toute étude de validation d'une nouvelle procédure, il est requis de déterminer la spécificité du test. Durant cet aspect de notre étude de validation un résultat positif au PSA fut obtenu pour des échantillons vaginaux sans sperme de même que pour une mousse contraceptive disponible dans le commerce. Cet article présente les résultats de validation et les guides d'interprétation lors de l'utilisation de ces tests au Centre des sciences judiciaires.
