Synthesis of DNA and Mitosis in Relation to Cell Differentiation in the Roots of Vicia Faba and Lactuca Sativa

SUMMARY

An analysis of mitotic index, labelling index and nuclear DNA content in differentiating cells of roots in Vicia faba and Lactuca sativa has shown a different behaviour of cell differentiation in the two species.

In Vicia faba, the mitotic activity is almost wholly restricted to the meristem (two apical mm.), whilst the DNA synthesis occurs also in the root areas in which cell differentiation and elongation are taking place. This makes the nuclear DNA content of differentiated cells 4C and, in some cases, 8C. In Lactuca sativa, DNA synthesis occurs only in the meristem, and the mitoses which take place at the base of the meristem exhaust the cells in the G2 (4C) phase of the mitotic cycle, so that the differentiated cells have a 2C nuclear DNA content.

RIASSUNTO

L'analisi dell'indice mitotico, dell'indice di marcatura e del contenuto nucleare di DNA, condotta in cellule in via di differenziazione nelle radici di Vicia faba e Lactuca sativa ha dimo strato un diverso comportamento delle due specie durante la differenziazione cellulare. In Vicia faba l'attività mitotica è per lo più limitata al meristema, mentre la sintesi del DNA si svolge anche in quelle zone dove hanno luogo la differenziazione e la distensione cellulare. Questo fatto porta il contenuto nucleare di DNA delle cellule differenziate sul valore 4C e in alcuni casi 8C.

In Lactuca sativa la sintesi del DNA è strettamente limitata al meristema e le mitosi che si svolgono alla base dello stesso, esauriscono completamente le cellule della fase G2 (4C) del ciclo mitotico, così che le cellule differenziate presentano soltanto contenuti 2C di DNA nucleare.
