Analysis of Nucleic Acid Bases in Photographic Gelatins by High-Performance Liquid Chromatography (on Free Bases and Combined Bases)

Abstract

A high-performance liquid chromatographic method was applied to the analysis of nucleic acid bases in photographic gelatins by a cation exchange resin. The sample photographic gelatin was hydrolyzed with hydrochloric acid. The contents of nucleic acid bases were calculated quantitatively from their calibration curves and the chromatograms of photographic gelatins. Highly restraining gelatins contained a relatively large amount of these bases, while lowly restraining or inert gelatins contained little of them. In highly restraining gelatins, the ratio of adenine to guanine was approximately 1: 1. It shows that highly restraining gelatins contained not only nucleic acid but also free nucleic acid bases and so on.

Résumé

Les bases constitutives des acides nucléiques contenus dans des gélatines photographiques ont été analysées par une technique très performante de chromatographie liquide sur une resine échangeuse de cations. Les échantillons de gélatine ont été hydrolysés en milieu chlorhydrique. Les teneurs de ces bases ont ete déterminées d’apres les chromatogrammes afferant aux gélatines photographiques en se réferant à des courbes d’étalonnage. Les gélatines fortement retardatrices contenaient une quantité relativement grande de ccs bases alors que les gélatines faiblement retardatrices ou inertes en contenaient peu. Dans les gélatines fortement retardatrices, les teneurs en adénine et en guanine étaient approximativement égales. Ce la montre que ces gélatines contenaient non seulement des acides nucléiques mais également des bases libres.

Zusammenfassung

Die Trennung von Purinbasen in photographischen Gelatinen wurde millels HPL (High-Performance Liquid hromatography) über Kationenaustauscher durchgeführt. Die zu untersuchende Gelatineprobe wurde dabei mit Salzsaure hydrolysiert. Der Gehalt an Purinbasen wurde durch Vergleich der hromatogramme mit den Kalibrierung kurven der reinen Basen quanlitativ bestimmt. Stark hemmende Gelatinen enthielten einen relativ hohen Anteil an Purinbasen wohingegen der Anteil in schwach hemmenden oder inerten Gelalinen gering war. In stark hemmenden Gelatinen waren Adenin und Guanin etwa zu gleichen Anteilen vorhanden. Es wurde weiterhin gefunden, das stark hemmende Gelalinen neben den komplellen Nucleinsauren auch freie Purinbasen enthalten.

Riassunto

Un metodo avanzato di cromatografia in fase liquida è stato applicato all’analisi delle basi degli acidi nucleinici delle gelatine fotografiche mediante una resina a scambio cationico. Il campione di gelatina è stato idrolizzato con acido c1oridrico. 11 contenuto delle basi degli acidi nucleinici e stato calcolato quantitativa mente dalle lore curve di taratura e dai cromatogrammi delle gelatine fotografiche. Le gelatine molto ritardate contengono quantitativi elevati di que te basi, mentre in que lie poco ritardate e inerti il contenuto è basso. Nelle gelatine molto rilardate, il rapporto adenine-guanina è quasi unitario. io significa che nelle gelatine molto ritardate vi è presente non soltanto acido nucleinico, rna anche basi libere e simili.
