On the Method for the Quantitative Separation of Nucleic Acid in Native Form from Photographic Gelatin

Abstract

A method for the quantitative isolation of nucleic acid from photographic gelatin was studied. The method consists of two steps; one is hydrolysis of the gelatin backbone in which a proteolytic enzyme free from nucleases is employed and the other is adsorption chromatography with DEAF Sephadex A–50. In application lo some photographic gelatins the nucleic acid was satisfactorily isolated in about 75%, overall yield.

Résumé

On a mis au point une technique permettant d’extraire quantitativement les acides nucléiques contenus dans les gélatines photographiques. Cette technique comporte deux étapes: hydrolyse du squelette protéique de la gélatine à l’aide d’un enzyme protéolytique exempt de nuclease et chromatographie d’adsorption sur résine DEAE Sephadex A 50. Appliquée à quelques gelatines photographiques, cette technique a permis d’isoler de façon satisfaisante les acides nucléiques avec un rendement global d’environ 75%.

Zusammenfassung

Es wurde ein Verfahren zur quantitativen Isolierung von Nukleinsäuren aus photographischen Gelatinen untersucht. Dabei wird die Gelatinekette zunächst mit Hilfe eines proteoly tischen Enzyms, das frei von Nukleasen ist, hydrolysiert und der Nukleinsäuregehalt anschliessend durch Adsorptions-Chromatographie mit DEAE Sephadex A-50 ermittelt. Bei der Anwendung des Verfahrens auf verschiedene photographische Gelatinen konnten etwa 75% des Gesamtnukleinsäuregehaltes isoliert werden.

Riassunto

E’ stato studiato un metodo per isolare quantitativamente l’acido nucleinico dalla gelatina fotografica. Questo metodo consiste di due fasi: la prima è l’idrolisi della catena della gelatina operata con un enzima protelitico privo di nucleasi; la seconda è l’adsorbimento cromatografico con DEAE Sephadex A-50. Applicandolo a diverse gelatine fotografiche si è ottenuto un isolamento soddisfacente dell’ac, nucleinico con una resa del 75%.
