Genetic Investigations with Dipodascus Uninucleatus

SUMMARY

Biochemical mutants have been isolated in Dipodascus uninucleatus. The sensitivity of the wild type or specific biochemical mutants to ultraviolet light (UV) is independent of the medium on which the cultures were grown up preparatory to irradiation or the plating medium following UV treatment. When vegetative cells and ascospores from two distinct biochemical mutants are mixed together on complex medium, suspensions of vegetative cells and ascospores can be obtained following incubation of which a fraction can grow on minimal medium. This prototrophic fraction of the suspension probably results from genetic recombination at meiosis rather than somatic diploid formation since the double auxotrophic recombinant expected from genetic recombination at meiosis has been recovered in one of the crosses.

The biochemical mutants investigated have little effect on the stereo-specific conversion of a precursor for the total in vitro synthesis of prostaglandin E1 (PGE1).
