Comparison of two culture media for primary isolation of Paracoccidioides brasiliensis from sputum

Abstract

An experimental study was undertaken to determine the value of 2 culture media for the primary isolation of Paracoccidioides brasiliensis from seeded sputum samples. It was found that yeast extract agar was superior to modified Sabouraud's agar as recovery rates were 96.8% for the former and 65·6% for the latter. Apparently such effectiveness was due not to the intrinsic nature of the medium itself but that the growth of Candida spp. was discouraged on yeast extract agar. Media poor in carbohydrates, such as yeast extract, should thus be employed when attempting to isolate dimorphic fungi from samples expected to contain Candida spp.

Se emprendió un estudio experimental con el fin de determinar la efectividad de dos medios de cultivo en el aislamiento primario del P. brasiliensis a partir de muestras de esputos. Se halló que el medio de extracto de levadura era superior al agar modificado de Sabouraud, permitiendo el aislamiento en 96.8% de los casos, en contraste con el 65·5% demostrado por el Sabouraud. La eficacia del extracto de levadura parece ser debida no a la composición intrínsica del medio mismo, sino al hecho de que tal sustrato no estimula el crecimiento de levaduras del género Candida. Se observó, igualmente, que el crecimiento de Candida ejerce efectos nocivos sobre el desarrollo inicial del P. brasiliensis, impidiendo su aislamiento cuando el número de colonias de Candida es abundante. Se sugiere que, cuando se intente aislar hogos dimórficos de muestras en las que se espera encotrar levaduras tipo Candida, se empleen medios pobres en carbohidratos, tales como el agar extracto de levadura.
