The formation of germ tubes by Candida albicans in various peptone media

Abstract

Seventy-nine isolates of Candida albicans and 33 of 5 other pathogenic species of Candida were cultured for 3 h at 37°C in 1·5% aqueous solution of 6 peptone media. While all isolates of C. albicans formed germ tubes, the other species did not. Some isolates of C. albicans formed germ tubes in 1·5% trypticase peptone and neopeptone, and all did so in phytone, bactopetone, polypeptone and proteose peptone at the same concentration. Subsequently, 140 isolates of C. albicans were inoculated at the rate of 106 organisms/ml into concentrations of bactopeptone from 0·5% to 2·5%, into distilled water and into undiluted sheep serum. The tendency to germ tube formation was greater in 1% than in 0·5% bactopeptone, and in both of these media it was greater than in other concentrations of bactopeptone or whole sheep serum. Germ tube formation also occurred to a very limited extent in distilled water. The significance of these findings is discussed.

Resumen

Setenta y nueve cepas de Candida albicans y 33 cepas de otras 5 especies patógenas de Candida fueron incubadas durante 3 horas a 37°C en solución acuosa al 1'5% de seis medios peptonados. Mientras que todas las cepas de C. albicans formaron tubos germinales, ninguna de las otras especies 10 hicieron. Algunas cepas de C. albicans formaron tubos germinales en peptona tripticasa y neopeptona al 1'5% y todas, a la misma concentración, en fitona, bactopeptona, polipeptona y peptona proteasa.

Posteriormente, 140 cepas de C. albicans fueron inoculadas en la cantidad de 1 × 106 organismos/ml en concentraciones de bactopeptona entre 0'5% y 2'5%, también en agua destilada y en suero ovino sin diluir. La tendencia a formar tubos germinales fue mayor en bactopeptona al 1% que al 0'5% y en esta, dos concentraciones mas que en otras de bactopeptona o en suero ovino. La formación de tubos germinales también se producía en agua destilada, aunque sólo en muy pequeña cantidad. El significado de estos hallazgos se discute en relación con la prueba de tubos germinales para C. albicans y los factores que pueden intervenir en esta prueba.
