Aspects of the dimorphism of Histoplasma farciminosum: A light and electron microscopic study

Abstract

Within 48h following the induction of mycelial to yeast-like phase conversion of Histoplasma farciminosum, randomly occurring hyphal cells were observed to contain multiple nuclei and markedly increased numbers of mitochondria. Yeast-like cells arose as buds from swollen tips of terminal hyphae, as sessile buds along the hyphae, and as buds from chlamydospores. Yeast-like cells were characterized by the presence of numerous buds over the surface of the mother cell. Bud scars were evident in the cell wall of the mother cell following abscission of the bud cell. Little similarity was noted between the fine structure of yeast-like H. farciminosum and that reported for H. capsulatum. The yeast-like cells of H. farciminosum underwent rapid transformation to the mycelial phase at 25°C. The hyphal cell wall originated from the inner layer of the cell wall of the yeast-like form. The cytoplasm of the hyphal cell usually contained a single nucleus, scattered mitochondria and occasional lipid storage bodies. Occasionally, Woronin bodies were observed at the septal pore.

Dans les 48 heures qui suivent l'induction de la transformation de la forme mycélienne d'Histoplasma farciminosum en forme levure, apparaissent au hasard, des cellules hyphales qui contiennent des noyaux multiples et un nombre nettement accru de mitochondries. Des cellules levuriformes naissent à partir de bourgeons d'apex enflés d'hyphes terminaux, de bourgeons sessiles le long des hyphes et de bourgeons de chlamydospores. Les cellules levuriformes sont caractérisées par la présence de nombreux noyaux sur toute la surface de la cellule mère. A la suite de la séparation du bourgeon et de la cellule mère, on observe dans la paroi de cette dernière une cicatrice de bourgeonnement évidente. On ne note que peu de ressemblance entre l'ultrastructure de la forme levure de H. farciminosum et celle rapportée pour H. capsulatum. A 25° C, les cellules levures de H. farciminosum subissent une rapide transformation en phase mycélienne. La paroi de la cellule hyphale nait de la couche interne de la paroi cellulaire de la forme levure. Le cytoplasme de la cellule hyphale contient habituellement un seul noyau, des mitochondries dispersées et, occasionnellement, des corps de réserve lipidique. On peut parfois observer des corps de Woronin au niveau du pore du septum.
