Ultrastructure of Coccidioides immitis after exposure to the imidazole antifungals miconazole and ketoconazole

Abstract

Scanning and transmission electron microscopy was performed on the various phases of Coccidioides immitis, exposed for different periods of time to the imidazole antifungals miconazole and ketoconazole.

The development of spherules into endospores, which takes place in cultures under normal growth conditions, was suppressed in the drug treated cultures.

Typical ultrastructural changes were localized at the cell periphery and in the vacuolar system. The drugs did induce changes in mature, resting endospore cultures and in cultures incubated statically at room temperature. Aerobically growing endospores were not susceptible to either drug. The transformation of arthroconidia into mycelium was fully prevented after treatment. Mycelial cells were most susceptible to the antifungals for necrosis was induced in a substantial part of the hyphae after exposure for 24 h.

Résumé

Les techniques de microscopie électronique à balayage ou par transmission ont été appliquées aux différentes phases de Coccidioides immitis, exposées pendant des périodes de temps variées au miconazole et au kétoconazole, dérivés d'imidazole.

La transformation des sphérules en endospores qui se produit dans des conditions normales de culture, est supprimée dans les cultures traitées.

Les modifications ultrastructurales typiques sont localisées à la périphérie de la cellule et dans le système vacuolaire. Les médicaments induisent des changements dans les cultures d'endospores mures et dans les cultures incubées, san agitation, à la température du laboratoire. Les endospores cultivées en aérobiose ne sont pas sensibles à ces médicaments. La transformation d'arthroconidies en mycélium est totalement arrêtée après traitement. Les cellules mycéliennes sont très sensibles aux antifongiques et une grande partie de l'hyphe se nécrose après un contact de 24 heures.
