Abstract
5-Fluorocytosine (5FC) inhibits the germination of the conidia of A. fumigatus. This is different from the lack of effect of 5FC on hyphal formation from blastospores of C. albicans because, unlike C. albicans, A. fumigatus requires DNA synthesis for germination, and this is inhibited by 5FC.
A modified fluorescent Feulgen method has been developed to stain fungal nuclei in the presence and absence of 5FC. This method has shown that hyphae treated with 5FC have fewer nuclei than untreated hyphae.
During hyphal growth, DNA synthesis and hyphal elongation in A. fumigatus are equally inhibited by 5FC. This is different from the differential effect of 5FC on C. albicans when hyphal elongation continues during an inhibition of DNA synthesis. The equal inhibition in A. fumigatus is clearly not unbalanced growth and so the action of 5FC on A. fumigatus is merely fungistatic.
Zusammenfassung
5-Fluorocytosin (5FC) verhindert das Sprossen von Conidien bie Aspergillus fumigatus. Diese Wirkung unterscheidet sich zur fehlenden Wirkung von 5FC auf die Keimschlauchbildung bei Candida albicans. Im Unterschied zu C. albicans, ist die DNS-Synthese bei A. fumigatus für das Sprossen notwendig. 5FC hemmt die DNS-Synthese.
Eine modifizierte fluoreszierende-Feulgen Methode wurde zur Färbung von Pilzkernen mit und ohne 5FC entwickelt. Es konnte damit bewiesen werden, dass 5FC-behandelte Hyphen weniger Kerne haben, als Kontroll-Hyphen.
Bei A. fumigatus hemmt 5FC im gleichen Mass die DNS-Synthese wie das Hyphen-Wachstum. Dies ist unterschiedlich zur Wirkung von 5FC auf C. albicans, wo sich das Hyphen-Wachstum auch bei Hemmung der DNS-Synthese fortsetzt. Die gleichzeitige Hemmung bei A. fumigatus kann nicht mit “unbalanced growth→d erklärt werden; die Wirkung von 5FC auf A. fumigatus ist deshalb nur fungistatisch und nicht fungizid.
