Infusion of pentagastrin was given intravenously to patients with peptic ulcer disease. The dose that gave the maximal acid output varied between 0.5 and 5.1 μg/kg/hr. Even small doses gave a pronounced increase in pepsin output and increasing doses gave a decreasing ratio of pepsin to acid. Treatment with optimal effective doses of 1-hyoscyamine in tablets with sustained release resulted in a decreased output of acid and pepsin in the basal period and with low doses of pentagastrin stimulation. With higher doses of pentagastrin no difference was seen. The increase in pepsin output during pentagastrin stimulation was more pronounced during treatment with 1-hyoscyamine than without.
