A method is described for the quantitative determination of bile acids in biological material. It is based on the specific enzymatic conversion of the 3α-hydroxyl group of the steroid molecule into a 3-keto-group, using a purified 3α-hydroxysteroid dehydrogenase and spectrophotometric determination of NADH. The individual major bile acids are measured after separation by thin-layer chromatography. The procedure of quantitation is specific, simple, rapid, and reliable.
