The effect of pentagastrin on the cyclic 3′,5′-adenosine monophosphate (cyclic AMP) content of human gastric mucosa was studied. Measurement of cyclic AMP was correlated with the acid secretory response. In 21 healthy subjects, following pentagastrin administration (6 µg/kg b.w., s.c.) gastric secretion increased significantly, whereas the cyclic AMP content of gastric mucosa did not exceed basal values. In 2 patients with elevated serum gastrin levels and excessive basal hyperchlorhydria, cyclic AMP contents of gastric mucosa were not different from those in control subjects. From these results, pentagastrin-stimulated peak gastric acid output in man is not associated with an accumulation of cyclic AMP in gastric mucosa.
