Abstract

A simple and rapid HPLC method has been developed for the determination of creatinine in urine.

Urine was diluted (1:50 v/v) with water and injected directly. The chromatographic separation was accomplished with butylamine-phosphoric acid buffer at a flow rate of 1.2 ml/min. The HPLC column was a cartridge packed with 3 μm Econosphere C18. The UV detection wavelength was 230 nm. The intra and inter assay coefficient of variation were less than 5.2%. The method was validated by comparing data on 232 urines with that obtained by the Jaffe method. It is suitable for routine determination of creatinine in urine as a measure of excretion rate, and is presently being used to improve the quality of the data from drug testing programs.
