РЕЗЮМЕ
Представлен краткий обзор современных представлений о строении и функции фибронектина—гликопротеина, входящего в группу клеточно-адгезивных белков. Обсуждается его роль в процессах эмбрионального развигия, регенерации, злокачественной трансформации, механнзмов клеточного прикрепления и адгезии и др.
Описана технология и опыт авторов в получении фибронектина из плазмы крови человека. Использован метод аффинной хроматографии на желатин-сефарозе 4 Б. Обсуждаются способы освобождания препарата от протеолитических энзимов. контрля его чистоты и биологической активности, а также создания оптимальных условий сохранения.
SUMMARY
Short view of the current scientific data about the structure and function of the fibronectin—a cell adhesion glycoprotein was made. Its role in the embryo development processes, regeneration, malignant transformation, mechanism of cell attachment and adhesion was discussed.
The authors describe the detailed technology of fibronectin purification from human plasma using the method of affinity chromatography on gelatin—sepharose 4B.
The ways for proteolytic enzymes inhibition and release, control of purity and biological activity of the fibronectin, as well as optimal conditions for storage was shown.
