РЕЗЮМЕ
Разработан колориметрический метод исследования глюкозоизомеразной активности с помощью 2-тиобарбитуровой кислоты, который отличается высокой специфичностью, достаточной точностью и чувствительностью (0,5 μМ фруктозы/ml). Исследована глюкозоизомеразная активность иммобилизованных нитактных клеток Streptomyces griseoflavus 1339 и других ферментных препаратов в условиях непрерывного перемещивания. Результаты исследования ферментной кинетики (влияние рН, температуры, концентрации субстрата, количества ферментного препарата и др.) показывают, что тиобарбитуровый метод подходит для аналитических целей. Быстрота и простота его выполнения позволяют осуществлять автоматизированный контроль за глюкозоизомеразным процессом в промыщленных условиях.
SUMMARY
Colorimetric method has been developed to determine glucoseisomerase activity using 2-thiobarbituric acid which exhibits high specifity, accuracy and sensitivity (i. e. 0.5μM fructose/ml). Studies were carried out on the glucoseisomerase activity of immobilized intact cells of Streptomyces griseoflavus 1339 as well as of other enzyme preparations under constant stirring. The results from the investigations on enzyme kinetics (i. e. effect of pH, temperature, substrate concentration, quantity of enzyme, etc.) show that the thiobarbituric method is suitable for analytical purposes. Its speed and simplicity allow application of automatic control of the glucoseisomerase process under industrial conditions.
