Abstract
A modified method for the analysis of porphyrin carboxylic acids in mink urine by high performance liquid chromatography was validated. The influence of instrument variables, such as flow rate and carry‐over, and analytical variables, such as percent recovery and repeatability, were assessed. The limit of detection for individual porphyrins was 10 fmol (20 μl injection) which represents 0.5 nM in mink urine. The repeatability of the method ranged from 7.0% to 10.0%. Study of the percent recovery revealed a method bias for heptacarboxyporphyrin and pentacarboxyporphyrin. This bias does not influence toxicological studies since reference values and a control group are used for the comparison.
