Beitrag zur differenzierung von haemophilus‐stämmen aus hühnern

Zusammenfassung

Für die serologische Untersuchung im Objektträger‐Aggluti‐nations‐Test (OA‐Test) standen 31 Feldisolate aus Hühnern zur Verfügung, von denen 23 der Species Haemophilus para‐gallinarum angehörten. Je ein H. paragallinarum‐Stamm des Serotyps A und des Serotyps B (Page et al. 1963) sowie ein weiterer serologisch nicht eingeordneter Stamm aus Kalifornien/ USA dienten als Referenzstämme. Außerdem wurde ein H. parainfluenzae‐Stamm iin die Untersuchungen einbezogen.

Bei den H. paragallinarum‐Isolaten waren im OA‐Test mit Anti‐seren von Kaninchen zweli verschiedene Serotypen zu unterscheiden. Zwischen den Serotypen traten Kreuzreaktionen auf, die durch Verdünnung oder Absorption der Antiseren ausgeschalten werden konnten. Von 23 H. paragallinarum‐lsolaten gehörten acht dem Serotyp A und 13 dem Serotyp B an. Zwei Isolate zeigten in 0.85% iger NaCl‐Lösung Spontanagglutination Von acht H.‐Isolaten, die kulturell und biochemisch eine gesonderte Gruppe darstellten, war bei sieben Isolaten auch serologisch ohne Schwierigkeüten eine Unterscheidung von den H. paragallinarum‐Stämmen möglich. Ein Stamm zeigte hingegen Spontanagglutination. Die übrigen sieben Isolate der Gruppe konnten aufgrund der Ergebnisse im OA‐Test in vier Serotypen eingeteilt werden, wobei drei Stämme dem Serotyp 1, zwei dem Typ 2 und jeweils ein Stamm dem Serotyp 3 und 4 angehörten. Zwischen den Serotypen wurden nur sehr schwache Kruzreak‐tionen beobachtet.

Der H. parainfluenzae‐Stamm war mit keinem der aus Hühnern isolierten und untersuchten Stämme serologisch identisch.

Summary

A total of 31 field isolates, 23 of them belonging to the H. paragalli‐narum species were examined serologically by means of the rapid slide agglutination test (RSA‐test). One H. paragallinarum strain of serotype A, one of serotype B, and one serologically unclassified strain from California were used as reference strains. In addition, one stralin of H. parainfluenzae was included in these studies. On the basis of the results of the RSA‐tests With rabbit antisera 2 distinct serotypes of H. paragallinarum were differentiated. Cross reactions were observed among the iserotypes which could be eliminated by dilution and absorption of the antisera. Eight of 23 H. paragallinarum isolates belonged to erotype A and 13 to serotype 6. Two isolates showed spontaneous agglutination in 0.85% saline. Seven of 8 other isolates of Haemophilus bacteria, representing a culturally and biochemically separate group, could be distinguished from H. paragallinarum strains without difficulty. One strain showed spontaneous agglutination. The other 7 isolates formed 4 distinct serotypes on the basis of the RSA‐tests, of which 3 isolates belonged to serotype 1, 2 to serotype 2, and one each to serotypes 3 and 4. Weak cross reactions were observed among these serotypes. No strains isolated from chickens included in this study were sero‐logically identical with the istrain of H. parainfluenzae.

Résumé

31 isolats naturels, dont 23 appartiennent á l'espèce H. paragallinarum ont été utilisés pour des études sérologiques au moyen de l'épreuve d'agglutination rapide sur lame (S.A.R.). Une souche de H. paragallinarum du sérotype A, une du sérotype B, et une non classée provenant de Californie, ont été employées comme souche de référence. De plus, une souche de H. parainfluenzae a été incluse dans la présente étude.

Sur la base des résultats des S.A.R. avec des sérums‐anti de lapin, deux sérotypes différents ont été différenciés dans les souches de H. paragallinarum. Des réactions croisées parmi les sérotypes opt été observées et ont pu être liminées par dilution et adsorption des serums‐anti. 8 des 23 lisolats de H. paragallinarum appartiennent.au sérotype A, et 13 au sérotype B. Deux isolats donnent une agglutination spontanée avec le soluté chloruré sodique à 0,85 p. 100. 7 des 8 autres isolats d'Haemophilus, représentant un groupe distinct, du point de vue cultural et biochimique, ont pu être différenciés sans difficulté des souches d’H. paragallinarum. Une a donné une agglutination spontanée. Les 7 autres forment 4 sérotypes différents d'après la S.A.R.: 3 sont du sérotype 1, 2 du sérotype 2, 1 du sérotype 3, et 1 du sérotype 4. Parmi eux, quelques aibles réactions croisées sont observées. Toutes les souches de cette étude se osnt révélées sérologiquement différentes de H. parainfluenzae.
