Summary
Enzyme linked immunosorbent assays (ELISAs) incorporating up to three different antigens for screening of avian sera for antibodies to several viruses or mycoplasma are described. A triple antigen test comprising Newcastle disease virus (NDV), infectious laryngotracheitis (ILT) virus and avian influenza (AI) antigens for screening sera normally negative for antibodies to these viruses, was shown to be as sensitive as the corresponding single antigen ELISA in detecting seroconversion in experimentally inoculated birds and was also as sensitive as the haemagglutination‐inhibition (HI) test for NDV and AI, and the serum neutralisation test for ILT virus. Sensitivity was also demonstrated by comparison of end‐points in serially diluted NDV, ILT or AI positive sera.
A Mycoplasma synoviae ELISA was shown to be as sensitive as HI test for detection of MS and M. gallisepticum (MG) antibodies in experimentally inoculated birds and in field sera, and this antigen combined with NDV detected antibodies to MG, MS and NDV with sensitivity equivalent to the HI test in each case. The advantages of using pooled ELISA preparation for screening large numbers of sera which are normally negative for the pathogens concerned are discussed.

Une réaction immuno‐enzymatique (ELISA), incorporant jusqu'à trois antigènes différents pour le testage de sérums aviaires, a été étudiée en vue de la recherche d'anticorps vis‐à‐vis de différents virus ou mycoplasmes. Un antigène triple, comprenant le virus de la maladie de Newcastle (NDV), le virus de la laryngotrachéite infectieuse (ILT) et les antigènes de l'influenza aviaire (AI), destiné à tester des sérums normalement négatifs en anticorps pour ces virus, s'est révélé être aussi sensible que les antigènes correspondants seuls en technique ELISA, pour déceler la séroconversion chez des oiseaux inoculés expérimentalement. Cet antigène s'est révélé aussi sensible que le test de l'inhibition de l'hémagglutination (HI) pour NDV et AI que la test de séroneutralisation pour le vírus ILT. La sensibilité a aussi été démontrée par comparaison des dilutions limites avec des sérums dilués positifs pour NDV, ILT ou AI.
Un test ELISA pour Mycoplasma synoviae (MS) s'est révélé aussi sensible que le HI‐test pour la détection des anticorps de MS et M. gallisepticum (MG) chez des oiseaux inoculés expérimentalement ou infectés naturellement. Cet antigène combiné avec NDV a permis de déceler des anticorps vis‐à‐vis de MG, MS et NDV avec une sensibilité équivalente au HI‐test dans chaque cas. Les avantages d'utilisation de préparations de mélanges avec la technique ELISA, pour le testage de grands nombres de sérums qui sont normalement négatifs pour les agents pathogènes concernés sont discutés.
Zusammenfassung
Es werden Enzymimmunosorbent‐Versuche (ELISA), die bis zu drei verschiedene Antigene inkorporiert haben, beschrieben. Sie sollen für den Nachweis von Antikörpern gegen verschiedene Viren und Mycoplasmen in aviären Seren benutzt werden, Ein Dreifachantigentest, der Antigene des Newcastlevirus (NDV), des Virus der Infektiösen Laryngotracheitis (ILT) und eines Aviären Influenzavirus (AI) enthielt, wurde für die Überprüfung von Seren verwandt, die normalerweise keine Antikörper gegen diese Viren aufwiesen. Dieser Test erwies sich für den Nachweis einer Seroconversion in experimentell inokulierten Tieren als ebenso empfindlich wie der entsprechende Einzelantigen‐ELISA und genau so wie der Haemagglutinationshemmungstest (HI) für NDV und AI und der Serumneutralisationstest für das ILT Virus. Die Empfindlichkeit war auch beim Vergleich der Endpunkte bei serienmäßiger Verdünnung positiver NDV‐, ILT‐ oder AI‐Seren feststellbar.
Ein Mycoplasma synoviae (MS) ELISA erwies sich für den Nachweis von MS‐ und M. gallisepticum (MG)‐Antikörpern in experimentell inokulierten Tieren und Feldseren als ebenso empfindlich wie der HI Test. In jedem Fall konnte mit diesem mit NDV kombinierten Antigen Antikörper gegen MG, MS und NDV mit der gleichen Empfindlichkeit nachgewiesen werden wie mit dem HI Test. Die Vorteile der Verwendung gepoolter ELISA Praeparationen bei Prüfung großer Serazahlen, die normalerweise bezüglich der betreffenden Pathogene negativ sind, werden diskutiert.
Resumen
Se describien métodos de enzimoinmunoenzayo (ELISAs) a los que se les incorpora hasta tres antígenos diferentes para realizar un muestreo de anticuerpos en el suero aviar de diversos virus o micoplasmas. Se demostró que el método con un antígeno triple que comprendía al virus de la enfermedad de Newcastle (NDV), al virus de la laringotraqueitis infecciosa (ILT) y a antígenos de la influenza aviar (AI) para hacer un muestreo de sueros generalmente negativos a la presencia de anticuerpos frente a estos virus era tan sensible como el ELISA correspondiente con un sólo antígeno, en la detección de una seroconversión en aves inoculadas experimentalmente, siendo también tan sensible como la prueba de la inhibición de la hemoaglutinación (HI) para NDV y AI y la de la seroneutralización para el virus ILT. Se demostró también la sensibilidad comparando la dilución final de los sueros positivos a NDV, ILT o AI diluidos seriademente.
El ELISA para el Mycoplasma synoviae (MS) fue tan sensible como la prueba de la HI en la detección de anticuerpos frente al MS y al M. gallisepticum (MG) tanto en aves inoculadas experimentalmente como en sueros de campo, siendo este antígeno junto con el de NDV capaces de detectar anticuerpos frente a MG, MS y NDV con una sensibilidad equivalente al método de HI por separado. Se discuten las ventajas de emplear una mezcla de antígenos en el método de ELISA para hacer muestreos de grandes cantidades de sueros que son generalmente negativos a dichos agentes patogenos.
