Fast Kinetic Studies by Fluorescence Correlation Spectroscopy of Pna-Dna Interactions

Abstract

The kinetics of the hybridization of fluorescently labeled Peptide nucleic acid (PNA) with its complementary single strand DNA templates and strand displacement from the same DNA duplexed with its 18-nucleotide(nt) long sequencing primer was measured. We used M13mp18(+)ssDNA as template and the PNA molecule was 18-nt long with the identical sequence as to that of the sequencing primer of the DNA template. Kinetic studies show that hybridization was approximately 100-fold faster than the oligonucleotide-displacement at room temperature.
