Cell disintegration for the purification of intracellular proteins

Results from recent studies in mechanical cell disintegration are presented with regard to stirrer design in high speed mills and valve design in high pressure homogenizers. Pretreatment of Bacillus species with lytic enzymes at low concentration is shown to improve the efficiency of wet milling for cell disruption. Data from a preliminary analysis of particle size of the cell debris generated by the different methods are discussed.
