Methodological aspects and potential of computer‐aided protein engineering

Abstract

The techniques used in computer‐aided protein engineering make a rational design of proteins with new properties possible. The potential as well as the currently still existing shortcomings of this methods are described. Activities during a typical protein design cycles are described. It is concluded that the development of the methodology described provides a huge step in protein engineering. Areas where further development is needed, and areas where this progress is accomplished, are identified.
