Equilibrium dialysis has been used to study the effects of 60Co γ-radiolysis on the carbohydrate binding sites of Con A. Reaction of e−aq and OH· with Con A is accompanied by a decrease in the number of binding sites. Modification of the binding site by (Br) is independent of the initial concentration of Br−. Reaction of (SCN)
with the protein, however, is dependent on the initial SCN− concentration. The results imply that tryptophan and possibly tyrosine are probably involved in the carbohydrate binding process.
