Characterization of 1.8 kb Hind III Fragment of Clostridium septicum Genomic DNA

A genomic library of Clostridium septicum (IVRI strain 51) was generated by cloning Hind III fragments of genomic DNA of C. septicum into the plasmid pUC18. Thirty-three recombinant clones were screened and a recombinant, plasmid (pSDS12) containing a 1.8 kb insert was selected for characterization by restriction mapping. The map was confirmed by probing the restriction digests of the insert with 1.8 kb insert labeled with Digoxigenin-d-UTP. This insert has a unique Nsi 1 site. This Nsi 1 site could facilitate further sub-cloning as preliminary studies indicate that this insert may show utility as a gene probe in the diagnosis of C. septicum infections.
