Clostridium septicum is known to produce the largest amount of DNase, which poses a challenge in preparation of DNA of good integrity and quality for enzymatic manipulations. A method was evolved using mercuric chloride and a high concentration of SDS for the preparation of genomic DNA of C. septicum, yields genomic DNA of good quality and integrity. This method, therefore, circumvents the earlier tedious purification procedures involving cesium chloride/Ethidium bromide density gradient centrifugation.
