7,7-DIMETHYLCEPHEMES SUBSTITUES EN POSITION 4 PAR DES GROUPEMENTS METHOXYMETHYLE, PIPERIDINOMETHYLE OU ACETOXYMETHYLE

Abstract

A 4-methyl-2-methylene-3,6-dihydro-2H-1,3-thiazine 1 is converted into 4-methoxymethyl-H7a, 4-piperidinomethyl-H7b or 4-acetoxymethymethylcephem 7c by two different pathways. Either monobromination of the methyl group bound to the four position is followed by methanolic nucleophilic substitution then by four steps conversion of the new 2-methylene-3,6-dihydro-2H-1,3-thiazine 3a into cephem. Either monobromination occurs at the methyl group substituted at the four position of a firstly synthetized 4-methylcephem 9 followed by nucleophilic substitution by piperidine or sodium acetate.

Une 4-methyl-2-méthylène-3,6-dihydro-2H-1,3-thiazine 1 est convertie en 4-méthoxyméthyl-H7a, 4-pipéridinométhyl-H7b ou 4-acétoxyméthylcéphème 7c par deux voies différentes. Soit monobromation du méthyle en position 4, substitution nucléophile par le méthanol en milieu basique, puis conversion de la 2-méthylène-3,6-dihydro-2H-1,3-thiazine 3a obtenue en céphème; soit, monobromation du méthyle en position 4 et substitution nécleophile par la pipéridine ou l'acétate de sodium sur un 4-méthylcéphème 9 pré formé.
