SPE-HPLC TECHNIQUES FOR SEPARATION AND IDENTIFICATION OF DOMPERIDONE IN HUMAN PLASMA

Abstract

This paper describes a simple, rapid, sensitive, and validated high performance liquid chromatographic (HPLC) method for the determination of domperidone in plasma. The analyte (domperidone) and the internal standard (celiprolol) were extracted from the plasma sample by solid phase extraction (SPE) method. The extraction was carried out using methanol at a flow rate of 1.0 mL/min. The HPLC separation was carried out on symmetry C18 column (150 mm × 0.46 mm, 5 µm). Phosphate buffer (50.0 mM, pH 3.0)-acetonitrile (80:20, v/v) was used as a mobile phase at a flow rate of 2.0 mL/min. Detection was carried out at 240 nm. The recoveries of domperidone from standard and plasma samples were 88.5% and 88.0%, respectively. The values of retention factor of domperidone were 2.45 and 2.36 in standard and plasma samples, respectively. The values of separation and resolution factors of domperidone with respect to celiprolol were 2.00 and 20.0, respectively. The detection limit of HPLC for domperidone and celiprolol were 1.0 and 1.2 µg/mL, respectively.
