Evaluation of the Merits of the New Method of Oviductus Ranae by HPLC-DAD

Abstract

In this study, a new method of evaluating the merits of Oviductus Ranae has been established, and HPLC was used to determine the contents of 4-cholesten-3-one, 3β-hydroxy-cholesta-5-en-7-one, and cholesterol in Oviductus Ranae at the same time. The determination was achieved on a Waters Xbridge-C18 (4.6 × 250 mm, 5 µm) column maintained at a temperature of 30°C, using methanol–water (90:10%, v/v) as the mobile phase with the flow rate of 1.0 mL/min and detected using UV-Visible detector at a UV wavelength of 248 nm. Detection was carried out at the wavelength of 220 nm and 240 nm with the sample volume of 10 uL. The 4-cholesten-3-one, 3β-hydroxy- cholesta-5-en-7-one, and cholesterol in Oviductus Ranae of the analytical samples represent good liner relations in the range of 0.0030150–0.010050 µg, 0.003042–0.010140 µg, and 0.6126–2.0420 µg, respectively. The average recovery of hree samples were 98.8%, RSD = 0.48% (n = 6); 99.4%, RSD = 1.51% (n = 6); 100.2%, RSD = 1.2% (n = 6), respectively, and showed good stability, precision, and repeatability. This method can determine the contents of 4-cholesten-3-one, 3β-hydroxy-cholesta-5-en-7-one, and cholesterol in Oviductus Ranae at the same time and was proven to be fast, accurate, and stable.
