Isolation of cadmium excluding mutants of arabidopsis thaliana using a vertical mesh transfer system and ICP‐MS

Abstract

Two cadmium hypertolerant mutants of Arabidopsis thaliana, cdhtl and cdht4, were isolated using the Vertical Mesh Transfer system (Murphy and Taiz, 1995). Both cdhtl and cdht4 were able to grow their roots > 2mm upon exposure to 200 μ? CdCl2. Exposure to 90 μ? CdCl2 inhibited the root growth of wild type Arabidopsis plants. Analysis of Cd accumulation and partitioning by Inductively Coupled Plasma Mass Spectroscopy (ICP‐MS) analysis indicated that cdhtl and cdht4 accumulated 56% less total Cd than the wild type Arabidopsis plants. ICP‐MS analysis showed a significant reduction in shoot accumulation of Cd by cdhtl and cdht4 in comparison to wild type Arabidopsis. ICP‐MS analysis of root tissue also showed a reduction in the accumulation of Cd by cdhtl and cdht4. Assays of cadmium accumulation using ICP‐MS indicate that cdhtl and cdht4 mutants are effective Cd‐excluders as compared to wild type Arabidopsis.
