Activités antivirale et antioxydante in vitro d'huiles essentielles de Thymus capitatus (L.) Hoffmans. & Link de Tunisie

Abstract

Three essential oil samples of Thymus capitatus (L.) Hoffmans. & Link were tested for cytotoxicity by MTT assay in Vero cells and antiviral activity against type 1 Herpes simplex virus (HSV1), Echovirus 11 (ECV11) and Adenovirus (ADV) using cytopathic effect (CPE) reduction essay. In order to determine the mode of antiviral action of the essential oil, thyme was added at different times to the cells or viruses during infection. Both HSV1 and ECV11 were significantly inhibited when virus were pretreated with the essential oil prior to adsorption, while ADV was affected after penetration into the host cell. In vitro antioxidant properties of the essential oils, obtained by DPPH and ABTS assays, were interesting compared to the synthetic antioxidant Trolox.

Résumé

La cytotoxicité de trois échantillons d'huiles essentielles de Thymus capitatus (L.) Hoffmans. & Link a été testée sur des cellules Vero selon la méthode au MTT et leur activité antivirale investiguée vis-à-vis de Herpes simplex virus 1 (HSV1), Echovirus 11 (ECV11) et Adénovirus (ADV). Les huiles essentielles ont été incubées avec les virus et les cellules suivant trois modèles et leurs actions examinées en se basant sur l'inhibition de l'effet cytopathique des virus (ECP). Ces huiles se sont montrées douées d'une activité antivirale contre l'HSV1 et l'ECV11, en inhibant directement le virus avant même d'être inoculé aux cellules, et contre l'ADV, en empêchant sa réplication au sein des cellules hôtes. Ensuite, nous avons étudié le pouvoir antiradicalaire des huiles essentielles par les méthodes DPPH et ABTS. Les résultats ont démontré des activités prometteuses proches de celle du Trolox, l'antioxydant de référence.
