Hairy Root Transformation in Turkish Chickpea (Cicer Arietinum L) Cultivars

ABSTRACT

Rooting of Cicer arietinum L (Chickpea) cultivars Gökçe, Akçin 91 and Izmir 92 was evaluated using wild type 15834 strain of Agrobacterium rhizogenes by direct inoculation and puncture inoculation with needle. Scoring was done after 6 weeks. Directly inoculated explants showed a callogenic mass instead of hairy roots. There was a considerable difference in the highest frequency of hairy root formation which was achieved on variety Gökçe, in the light (16.67%) and dark (63.33%) after puncture inoculation. Mean transformation frequency ranged from 5 to 80%. Results show a significant difference for transformation from explants cultured in the light or dark.
