High Resolution of in Situ Hybridization in Plants' Mitotic Protoplasts

ABSTRACT

Since chromosome banding techniques were introduced into plant cytogenetics, many C-banding, N-banding and Q-banding techniques have been developed. But, these techniques only produce simple banding patterns in plant chromosomes. Based on the high resolution of in situ hybridization (FISH and GISH) in animals and human, we developed a new procedure which could yield high resolution in plant chromosomes.

The purpose of this article is to survey briefly a new technique that has generated new perspectives in a current molecular cytogenetical analysis. To obtain more metaphase cells with high quality for in situ hybridization, we suggested the use of protoplast of plants as an effective method.
