Interference of Some Modulators of Protein Phosphorylation and Ion Transport with Lactoferrin Stimulatory Effect on Erythrocyte Glycolysis

ABSTRACT

The aim of the present study was to test the ability of Lf to participate in the cell signals involved in the glycolysis control. We tested the Lf's effect on the lactate generation in the presence of some modulators of the protein phosphorylation (protein kinase and protein phosphatase inhibitors) and some modulators of the ion transport (calmodulin, W-7, amiloride and ouabain). Statistically reliable decrease of the Lf's stimulatory effect was found in the presence of wortmannin (with 45%, p<0.001), caffeine (with 18%, p<0.05), calmodulin (CaM) (with 22%, p<0.005), amiloride and ouabain (with 42% and 35%, resp., p<0.001). W-7, a CaM antagonist, enhanced the stimulatory effect of Lf with 42% (p<0.001). The PKC inhibitors did not change the effect of Lf. Our present results showed that Lf probably utilizes PI3К - and MAPK- dependent signals and is involved in cross-talks with CaM signaling.
