Investigation of DIA-1 Gene Polymorphism in Different Drosophila Species and Drosophila Melanogaster Stock Lines

ABSTRACT

The product of Drosophila melanogaster EG:22E5.5 or CG4199 (accession number O77266, Q9W529) from Berkely Drosophila Genome Project (BDGP) gene is Diaphorase-1, a NADH-dependent oxidoreductase containing NADH-binding domain and Rieske-type [2Fe-2S] cluster. The Dia-1 gene polymorphism was investigated in seven Drosophila species and ten mutant Drosophila melanogaster stock lines by PCR-SSCP analysis. Using appropriate primers we obtained 380 bp and 278 bp PCR products from different areas in the third exon of Dia-1 gene. These products were analysed by SSCP-electrophoresis followed by silver staining for band visualization. Conformation polymorphism was observed in Drosophila simulans and in all mutant Drosophila melanogaster stock lines.
