In-situ Production and Collection of Bacterial Cellulose on Jute and Flax Mats by Static Cultivation

ABSTRACT

In this study, jute and flax mats were used as a support material to deposit bacterial cellulose (BC) on its surface produced by Gluconacetobacter liquefaciens (MTCC 3135) through fermentation process under static condition. Amount of BC produced during the study indicates that type of mats as support material affects the production of BC at the air-liquid interface. About 2.45 g/l and 1.85 g/l of BC was produced in presence of jute and flax mats as support, respectively. Neat BC obtained under static cultivation was found to be brittle in nature. In order to prevent brittleness in BC, mat hanging method has been proposed to deposit BC directly on the surface of the mat. FTIR peaks obtained at 3337 and 3350 cm−1 as well as at 1647 and 1654 cm−1 indicated the presence of cellulose in form of BC on both the mats. Microscopic observation, water uptake behavior and antibacterial studies of neat mats, unadhered BC and BC deposited on mats were also carried out and the results were compared. The proposed in-situ method of collecting BC on the cellulosic mats will help in fabrication of biocomposite sheets that can be further explored in packaging, medical and biomedical applications.

本研究以黄麻和亚麻垫为支撑材料,在静态条件下,将液化葡萄糖酸杆菌(MTCC3135)发酵产生的细菌纤维素(BC)沉积在其表面. 在研究过程中产生的BC量表明,作为支撑材料的垫子类型影响气液界面BC的产生. 以黄麻和亚麻为载体,BC的产率分别为2.45g/l和1.85g/l. 在静态培养下获得的纯BC在性质上是易碎的. 为了防止BC的脆性,提出了挂网法将BC直接沉积在铺层表面. 在3337和3350cm-1以及1647和1654cm-1处获得的FTIR峰表明两种垫上都存在以BC形式存在的纤维素. 本文还对纯席、未附着的BC和沉积在席上的BC进行了显微观察、吸水行为和抗菌性能研究,并对结果进行了比较. 本文提出的在纤维素垫上原位收集BC的方法将有助于制备生物复合材料片,在包装、医疗和生物医学等领域有着广阔的应用前景.
